SYNTHESIS OF HYDROPHOBICALLY AND ELECTROSTATICALLY MODIFIED POLYACRYLAMIDES AND THEIR CATALYTIC EFFECTS ON THE UNIMOLECULAR DECARBOXYLATION OF 6-NITROBENZISOXAZOLE-3-CARBOXYLATE ANION

A series of hydrophobically and electrostatically modified polyacrylamides (Copol(AM-C12)) has been synthesized by radical-initiated copolymerization of acrylamide with n-dodecylmethyldiallylammonium bromide as the hydrophobe in aqueous solution using ammonium persulfate as the initiator. The formation of hydrophobic microdomains of the copolymers was revealed by large hypsochromic shifts of the long-wavelength absorption band of the solvatochromic probe Methyl Orange, noncovalently bound to the macromolecule. It was found that the microdomains formed by these copolymers in aqueous solution are more hydrophobic than those of the cationic polysoaps poly(alkylmethyldiallylammonium halides) containing the same n-dodecyl groups as the side chains as a result of the reduced electrostatic repulsions at the periphery of the microdomains. The reduced cationic character of the copolymers Copol(AM-C12) most likely also accounts for the observation that the anionic dye Methyl Orange does not; induce microdomain formation in aqueous solution. The effect of the hydrophobically and electrostatically modified polyacrylamides on the unimolecular decarboxylation of 6-nitrobenzisoxazole-3-carboxylate anion (6-NBIC) has been investigated in aqueous solutions at pH 11.3 and 30 degrees C. It is suggested that the relatively modest catalytic effects induced by Copol(AM-C12) should be ascribed to hydrogen-bond stabilization of the initial state by NH groups in the macromolecules. The decarboxylation rates of 6-NBIC at binding sites in hydrophobic microdomains increase with increasing n-dodecyl group content in the copolymers.

Compounds with strong absorptions in the ultraviolet (UV) region of the spectrum, particularly the UVA and UVB, have seen much interest as UV screeners or absorbers in a wide variety of commercial products. A series of benzisoxazole 2-oxides have been synthesized and characterized by UV-vis spectroscopy. A number of derivatives have been shown to posses moderate to strong molar absorption coefficients in the UVB range (ca. 300nm), the strongest being those derived from benzophenones. Three other derivatives containing additional electron withdrawing groups showed strong molar absorption coefficients in the UVA (ca. 340nm). Solvent effects on the parent derivatives show changes in the molar absorption coefficients with little changes in the max values. Preliminary studies of these compounds as potential additives to prevent photooxidation of polystyrene showed considerable inhibition of polymer degradation with the parent unsubstituted benzisoxazole 2-oxide compounds being the most effective. This difference is reflected in the more pronounced synergism in critical aggregation concentration and catalytic efficiency of the DTAB/SDS mixture and in the phase behavior of the mixtures. CTAB and SHS can be mixed in a 1:1 ratio without precipitation, forming both small, unilamellar and large, multilamellar vesicles. In DTAB/SDS mixtures, however, precipitation of the catanionic surfactant occurs for a mole fraction of DTAB (x) between 0.3 and 0.7, while both vesicles and large bilayer fragments are formed for x = 0.8. The excess of DTAB in the x = 0.8 mixture results in the solubilization of the Vesicles by DTAB micelles close to the cmc of pure DTAB. A network of interconnected threadlike cylindrical micelles was found as an intermediate stage of aggregation between the vesicles and the mixed micelles. These cylindrical micelles are formed exclusively on further increasing the surfactant concentration.

Research and development of zonisamide, a new type of antiepileptic drug

Zonisamide (1,2-benzisoxazole-3-methanesulfonamide, AD-810) is a broad spectrum antiepileptic drug which has been launched in Japan and South Korea. It lacks the ureide structure included in most of the existing antiepileptic drugs. Zonisamide was synthesized by the sulfonation and the successive amination of 1,2-benzisoxazole-3-acetic acid in a very poor yield. After several efforts to optimaize the compound, zonisamide was selected based on the balance of the efficacy and safety. The yield was greatly improved by the development of new synthetic routes. Zonisamide suppressed maximal electroshock seizures in mice, rats, rabbits and dogs. Its therapeutic plasma concentration range between anticonvulsant and neurotoxic effects was much wider than that of the existing antiepileptic drugs. In electroencephalographic studies on animal models of epilepsy, zonisamide, like phenytoin and carbamazepine, restricted the spread or propagation of seizures and, like sodium valproate, it suppressed the epileptogenic focus activity. Zonisamide was effective in several kindling models. In clinical studies, zonisamide exerted the efficacy against partial seizures (simple, complex, secondarily generalized seizures and some generalized seizures (tonic-clonic, tonic, atypical absence seizures) that were comparable to that of carbamazepine and sodium valproate, respectively. Zonisamide was also effective in monotherapy. The adverse effects related with zonisamide were mainly drowsiness, ataxia, loss of appetite and gastrointestinal symptoms. Serious adverse effects which may be life-threatening have not been reported.

X-Ray Structures of the LXR alpha LBD in Its Homodimeric Form and Implications for Heterodimer Signaling

Liver X receptors (LXRs) are nuclear receptors that are central regulators of cholesterol homeostasis, and synthetic LXR agonists have shown promise as promoters of reverse cholesterol transport and anti-inflammatory agents. Here, we present three X-ray structures of three different agonists bound to the ligand binding domain of LXR alpha. These compounds are GW3965, F(3)methylAA, and a benzisoxazole urea, and we show that these diverse chemical scaffolds address common structural themes, leading to high binding affinity for LXR. Our structures show the LXR ligand binding domain in its homodimeric form, an arrangement previously thought to be stereochemically difficult. A comparison with existing structures of the LXR beta homodimer and LXR alpha:RXR (retinoid X receptor) heterodimers explains differences in dimer affinity and leads us to propose a model for allosteric activation in nuclear receptor dimers, in which an unactivated RXR partner provides an inhibitory tail wrap to the cofactor binding pocket of LXR. Although possible usefulness of non-selective monoamine oxidase (MAO) inhibitors for Parkinson’s disease therapy has been suggested in the literature, MAO inhibitors whose inhibition is reversible and have dual action to both MAO-A and -B subtypes is not available yet. Subtype selectivity and reversibility of a series of novel MAO inhibitors, 3-(2-aminoethoxy)-1,2-benzisoxazole derivatives, were studied. Several dual MAO inhibitors, which inhibit both MAO-A and -B, were obtained. When administered to mice, their effects were generally reversible. Among the derivatives, RS-1636 and RS-1653 had much longer duration of brain MAO-B inhibition than that of MAO-A. In vitro, the inhibited MAO-A activity by these compounds was partially recovered by buffer change at 4degreesC, while little MAO-B activity was recovered. Although it is not fully elucidated yet, the reversibility of these inhibitors is probably determined primarily by this dissociation profile. This unique differential reversibility indicates that optimization of the balance of actions can be achieved by differentiating reversibility to each target molecule.

Multi-pharmacophore Approach to Bio-therapeutics: Piperazine Bridged Pseudo-peptidic Urea/Thiourea Derivatives as Anti-oxidant Agents

In an attempt to develop synergetic, potential and selective anti-oxidant agents, a series of novel piperazine bridged pseudo-peptidic urea/thiourea derivatives (5a-x) are designed by molecular docking study, synthesized and evaluated for their in-vitro anti-oxidant activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and dimethyl-4-phenylenediamine (DMPD) methods. The results revealed that compounds 5q, 5s, 5w and 5x are good anti-oxidant agents. Amongst, compounds with aromatic side chain, thioureal fragment with electron donating group-substitution, particularly, 5s and 5w are the most active members exhibiting remarkable anti-oxidant property. The docking results are complementary to the experimental results. The correlation study revealed that compounds derived from phenylalanine and tryptophan containing electron donating group are excellent antioxidants. [GRAPHICS] .

The catalytic antibody 21D8 efficiently catalyzes the decarboxylation of a substituted 3-carboxybenzisoxazole-a simple, unimolecular reaction that is not susceptible to general acid/base catalysis but that is highly sensitive to the microenvironment. The transition-state structure of this decarboxylation reaction has been probed by measuring the carbon kinetic isotope effect for the uncatalyzed decarboxylation in water and for the dioxane-accelerated and antibody-catalyzed reactions. The isotope effect for the decarboxylation of 5-nitro-3-carboxybenzisoxazole is k12/k13 = 1.046 in aqueous buffer at 20-degrees-C and 1.048 for the antibody-catalyzed reaction. With increasing dioxane in the reaction medium, the rate of the spontaneous decarboxylation increases by almost four orders of magnitude, whereas the isotope effect displays only a slight, progressive decrease to k12/k13 = 1.043 in 100% dioxane. Together, these results indicate that the structure of the transition state undergoes very little change in spite of > 10(4)-fold increases in the rate of the reaction caused by solvation of the substrate by organic solvents or the low dielectric environment of the antibody active site.
