Parkinson’s disease (PD) is a neurodegenerative disorder caused by the loss of dopaminergic neurons in the midbrain and shows motor dysfunctions. Zonisamide (ZNS, 1,2-benzisoxazole-3-methanesulfonamide), which was originally developed as an antiepileptic drug, was also found to have beneficial effects on motor symptoms in PD. In the current study, we have investigated the behavioral and physiological effects of ZNS on L-DOPA-induced dyskinesia (LID) in PD model mice. Chronic administration of L-DOPA plus ZNS in PD model mice was shown to increase the duration and severity of LID compared with PD model mice that were treated with L-DOPA alone. To elucidate the neural mechanism of the effects of ZNS on LID, we examined neuronal activity in the output nuclei of the basal ganglia, i.e., the substantia nigra pars reticulata (SNr). Chronic administration of L-DOPA plus ZNS in PD mice decreased the firing rate in the SNr while they showed apparent LID. In addition, chronic treatment of L-DOPA plus ZNS in PD mice changed cortically evoked responses in the SNr during LID. In the control state, motor cortical stimulation induces the triphasic response composed of early excitation, inhibition, and late excitation. In contrast, L-DOPA plus ZNS-treated PD mice showed longer inhibition and reduced late excitation. Previous studies proposed that inhibition in the SNr is derived from the direct pathway and releases movements, and that late excitation is derived from the indirect pathway and stops movements. These changes of the direct and indirect pathways possibly underlie the effects of ZNS on LID.

Upon reaction with DCP, bathochromic shift from similar to 463 to similar to 532 nm was observed due to the inhibition of PET from oxime-hydroxyl to the triphenyl amine moiety and cyclization to the benzisoxazole. The transformation from oxime to benzisoxazole was confirmed by NMR and HRMS data. The probe TPOD can be applied on a portable and cost effective test kits where in presence of DCP vapour dramatic color and fluorescence change were observed within 30 s. Furthermore, the probe can efficiently detect DCP in presence of other nerve agents and electrophile and the detection limit is 0.14 mu M. Moreover to evaluate the sensing mechanism we have prepared a control compound TPD whose crystal structure and non covalent interactions have been incorporated. To achieve the on-site application of TPOD we have performed an experiment in soil sample to detect trace amount of DCP. Moreover the probe can be useful in cellular imaging of DCP containing bio-samples. Thus the probe TPOD is an auspicious, instant and on site monitoring ratiometric fluorescent probe for selective DCP detection in presence of other interfering nerve agents.

We have synthesized 14 N-phenylurea derivatives, differing in the heterocyclic portion linked in N’-position, and tested their cytokinin-like activity. Results obtained indicate that the N-phenyl-N’-1,3,4-thiadiazol-2-ylurea (compound 4), an isomer of N-phenyl-N’- 1,2,3-thiadiazol-5-ylurea (thidiazuron, TDZ), in the absence of auxin induces shoot regeneration in the 34,2% of the explants cultured; the N-plienyl-N’-(3-chloro-1,2-benzisothiazol-7-yl) urea (compound 10), structurally different from TDZ, in the absence of auxin induces shoot regeneration in the 25,9% of explants, significantly lower than that of TDZ (68,8%). N-phenyl-N’-benzothiazol-6-ylurea (compound 13), structurally different from TDZ, in the absence of auxin induces the 99,5% of shoot regeneration, significantly different from that of the other substances. The addition of auxin in the cotyledon regeneration assay reduces the differences. The compound 13 could be considered a new phenylurea derivative with a highly specific cytokinin-like activity.
