Herein, we report spiropyrimidinetriones (SPTs) incorporating N-linked azole substituents on a benzisoxazole scaffold with improved Gram-positive antibacterial activity relative to previously described analogues. SPTs have an unusual spirocyclic architecture and represent a new antibacterial class of bacterial DNA gyrase and topoisomerase IV inhibitors. Purpose: Zonisamide (ZNS) and acetazolamide (AZM) are two antiepileptic drugs (AEDs) that differ in clinical efficacy. To elucidate the mechanisms of action of these compounds, we investigated their therapeutic and prophylactic effects in rats by using a kindling model of partial epilepsy. Methods: Electrodes were implanted into the left amygdala of adult male Wistar rats. The animals were stimulated at the afterdischarge threshold until Five stage 5 seizures were induced. The generalized seizure threshold was then determined. Therapeutic effects were examined in rats manifesting successive convulsions with near-threshold stimulation. To test prophylactic effects, drugs were administered intraperitoneally before daily kindling stimulation until the animal had a stage 5 seizure or reached day 18. Results: ZNS (10-40 mg/kg; n = 6) suppressed kindled seizures in a dose-dependent manner. Repeated administration for 7 days produced tolerance to anticonvulsive effects. AZM (25-200 mg/kg; n = 7) showed limited therapeutic effect, alleviating only the clonic convulsion in stage 5 seizures and reducing afterdischarge duration. Secondary generalization was not significantly suppressed during repeated treatment (50-200 mg/kg; n = 6). ZNS, 25 or 40 mg/kg (n = 8), significantly retarded seizure development; 15.0 or 17.0 daily stimulations were required to produce a stage 5 seizure. AZM, 50-200 mg/ kg (n = 6), also retarded seizure development, with 14.0-14.8 stimulations required. Conclusions: ZNS exhibited modest therapeutic and prophylactic effects, whereas AZM showed mainly prophylactic effects. Hypotheses are presented that may explain the mechanisms of action of these drugs.

Zonisamide (1,2-benzisoxazole-3-methanesulfonamide) was metabolized to its reductive product, 2-sulfamoylacetylphenol, in rat liver microsomes under anaerobic conditions. These results suggest that phenobarbital- and pregnenolone 16-alpha-carbonitrile-inducible form(s) of cytochrome P-450 is responsible for the reductive metabolism of zonisamide to 2-sulfamoylacetylphenol in rat liver microsomes.

Zonisamide is a synthetic 1,2-benzisoxazole-3-methanesulfonamide with anticonvulsant properties. Transition metal-catalyzed borylative difunctionalization of alkenes, which can simultaneously introduce C-B, C-C or C-X bonds, could directly construct highly functionalized organoboron in one step. Among these reactions, copper catalyzed enantioselective aminoboration of styrenes is an efficient approach to generate enantioriched beta-aminoboronate which is a class of useful chiral compounds. In this work, employing styrenes as substrates, 1,2-berrzisoxazole as an electrophilic primary amine source, bis(pinacolato)diboron (B(2)pin(2)) as boron source and LiOCH3 as base, an enantioselective Cu-catalyzed aminoboration of styrenes by using a chiral sulfoxide-phosphine (SOP) ligand was developed, and a board range of chiral beta-aminoalkylboranes, which could be readily converted to a class of valuable beta-hydroxylalkylamines, were accessed with high yields and ee values. A general procedure for this aminoboration of styrenes is described in the following: in a glove box, CuI (0.05 mmol), chiral sulfoxide phosphine ligand L1 (0.06 mmol), and 2 mL of anhydrous tetrahvdrofuran were added into a flame-dried tube. The resulting mixture was stirred at room temperature for 30 min. then bis(pinacolato)diboron (B(2)pin(2)) (0.75 mmol), LiOCH3 (1.25 mmol), styrene 1 (0.5 nunol), 1,2-benzisoxazole (0.75 mmol) and another 2 mL of THE were added into the reaction system in sequence. The reaction tube was removed out from the glove box and stirred at 20 degrees C for 12 h. After the reaction was finished, the NMR yield was firstly determined with dimethyl terephthalate (9.7 mg, 0.05 mmol) as internal standard, then, the crude product was recovered and purified with a preparative TLC which was alkalized with triethylamine to give the desired beta-aminoboronates in moderate to good yields (47%similar to 84%) and enantioselectivities (81%similar to 99%). To demonstrate the utility of this reaction, beta-boronate primary amine could be easily obtained by removing the Schiff base group of beta-aminoboronate 3 under the methanol solution of hydroxylamine hydrochloride, which could be further oxidized to give corresponding chiral beta-amino alcohol in moderate yield (48%).
