Rather efficient catalysis of the decomposition of 5-nitro-benzisoxazole to the cyanophenol was observed with antibodies elicited against a cationic hapten structurally unrelated to the benzisoxazole substrate. The rate enhancement by the most active antibody is better than 10(4) and the reaction is catalyzed by a carboxylate group associated with a hydrophobic binding site. (C) 1997 Elsevier Science Ltd.

4-{4-[4-Tetrahydrofuran-3-yloxy)-benzo[d]isoxazol-3-yloxymethyl]-piperidin-1-ylmethyl}-tetrahydropyran-4-ol (PF-4995274, TBPT) is a new agent that is a partial agonist of the human serotonin-4 (5-HT4) receptor and is under investigation for neurological disorders. Metabolism of TBPT was examined in vitro in human liver microsomes and human hepatocytes. Metabolites were also identified in the plasma of healthy human subjects in a phase 1 clinical study. Human-derived metabolite profiles were compared with corresponding profiles obtained in laboratory animal species. There were two major routes of metabolism in vitro: N-dealkylation of the methyltetrahydropyran moiety (M1) and hydroxylation at the seven position of the benzisoxazole moiety (M4). These were also observed in human plasma; however, in that matrix, the major metabolite was an unusual cyclized oxazolidine entity (M2). M2 was proposed to be formed via generation of an intermediate 4. iminium ion on the piperidine ring followed by spontaneous cyclization by attack of the beta-hydroxyl substituent of the tetrahydropyran ring to form a cyclized oxazolidine product. An authentic standard of the metabolite was generated using a methylene-blue-sensitized photochemical oxidation reaction as well as microbial transformation. Further investigation of this metabolite showed that it also possessed 5-HT4 agonism activity similar to the parent. The metabolite was 150-fold more highly protein bound in human plasma than TBPT, which is consistent with its presence as a major circulating metabolite while being only a minor metabolite in in vitro systems. Overall, this illustrates the importance of understanding the complex dispositional properties of a pharmacologically active metabolite. (C) 2013 Wiley Periodicals, Inc.

-2-Hydroxybenzaldoximes are reduced under mild conditions of ammonium formate/Pd-C in methanol to give secondary amines. Benzisoxazoles react under the same mild conditions to give the same products. A possible mechanism is suggested, involving the intermediacy of the benzisoxazole in the oxime conversion. (C) 2003 Elsevier Ltd. All rights reserved.

Zonisamide (ZNS; 3-sulfamoylmethyl-1,2-benzisoxazole), as one of the newer antiepileptic drugs, has been demonstrated its broad-spectrum clinical efficacy on various neuropsychiatric disorders. However, little is known regarding the mechanism of ZNS actions on ion currents in neurons. We thus investigated its effect on ion currents in differentiated hippocampal 19-7 cells. In whole-cell configuration of patch-clamp technology, the ZNS (30 mu M) reversibly increased the amplitude of K+ outward currents, and paxilline (1 mu M) was effective in suppressing the ZNS-induced increase of K+ outward currents. In inside-out configuration, ZNS (30 mu M) applied to the intracellular face of the membrane did not alter single-channel conductance; however, it did enhance the activity of large-conductance Ca2+-activated K+ (BKCa) channels primarily by decreasing mean closed time. In addition, the EC50 value for ZNS-stimulated BK Ca channels was 34 mu M. This drug caused a left shift in the activation curve of BK Ca channels, with no change in the gating charge of these channels. Moreover, ZNS at a concentration greater than 100 mu M also reduced the amplitude of A-type K+ current in these cells. A simulation modeling based on hippocampal CA3 pyramidal neurons (Pinsky-Rinzel model) was also analyzed to investigate the inhibitory effect of ZNS on the firing of simulated action potentials. Taken together, this study suggests that, in hippocampal neurons during the exposure to ZNS, the ZNS-mediated effects on BK Ca channels and A-type K+ current could be potential mechanisms through which it affects neuronal excitability.

New explortion of Sodium succinate hexahydrate

If you are hungry for even more, make sure to check my other article about 6106-21-4, Name: Sodium succinate hexahydrate.

Chemistry can be defined as the study of matter and the changes it undergoes. You’ll sometimes hear it called the central science because it is the connection between physics and all the other sciences, starting with biology. 6106-21-4, Name is Sodium succinate hexahydrate, molecular formula is , belongs to benzisoxazole compound. In a document, author is Millan, MJ, Name: Sodium succinate hexahydrate.

The novel benzodioxopyrrolidine (S-16924) displays a clozapine-like profile of interaction with multiple monoaminergic receptors, in addition to potent agonist activity at serotonin (5-HT)(1A) receptors. S-16924 (2.5 mg/kg i.p.) and clozapine (5.0 mg/kg i.p.) generated robust discriminative stimuli (DS) and displayed full mutual generalization. The D-4 antagonists L-745,870 and S-18126, the D-1/D-5 antagonist SCH-39166, and the D-3 antagonist S-14297 showed at most partial generalization to S-16924 and clozapine. The D-2/D-3 antagonist raclopride fully generalized to S-16924, but only partially generalized to clozapine. The 5-HT2A antagonist MDL-100,907 fully generalized to S-16924 and two further 5-HT2A antagonists, fananserin and SR-46349, showed partial generalization. However, MDL-100,907, fananserin, and SR-46349 showed less pronounced generalization to clozapine. Similarly, the 5-HT2C antagonists SB-200,646 and SB-206,553 more markedly generalized to S-16924 than to clozapine. The 5-HT1A receptor agonist (+/-)-8-dihydroxy-2-(di-n-propylamino) tetralin generalized fully to S-16924 but not to clozapine. Full generalization was obtained to both S-16924 and clozapine for the clozapine congeners, olanzapine and quetiapine. In distinction, the benzisoxazole, risperidone, and the phenylindole, sertindole, weakly generalized to S-16924 and clozapine. However, the benzisoxazole ziprasidone, which possesses 5-HT1A agonist properties, generalized fully to S-16924 but not to clozapine. Finally, the muscarinic antagonist scopolamine generalized fully to clozapine and partially to S-16924. In conclusion, S-16924 and clozapine display both communalities and differences in their compound DS; this likely reflects their respective complex patterns of interaction with multiple monoaminergic receptors. Although no specific receptor was identified as underlying the clozapine DS, 5-HT1A agonist as well as D-2 and 5-HT2A/2C antagonist properties contribute to the S-16924 DS.

Zirconocene alkyne complexes Cp(2)Zr(L)(Me(3)SiC(2)R) (L = Py, THF; R = SiMe(3), tBu) react with heterocyclic compounds like benzoxazole and related thiazoles to yield ring-expanded adducts Cp(2)Zr-C(SiMe(3))=C(R)-CH=N-o-C6H4-X (1-3) and Cp(2)Zr-C(SiMe(3))=C(SiMe(3))-CH=N-C(R’)=C(R’)-X (R’ = Me, H) (4, 5) by formal C-X (X = O, S) bond cleavage and coupling with the coordinated alkyne. In the case of benzisoxazole, the alkyne is not coupled but eliminated, and with ring-enlargement of the benzisoxazole a N-bridged dimer [Cp(2)Zr-N=CH-o-C6H4-O](2) (6) is formed.
