Interspecific somatic hybrid plants were obtained by symmetrical electrofusion of mesophyll protoplasts of Medicago sativa with callus protoplasts of Medicago arborea. Rapid and High-Yield Electrosynthesis of Benzisoxazole and Some Derivatives

Cyclic voltammetry and controlled-potential (bulk) electrolysis have been used to explore the electrochemical reduction of o-nitrobenzaldehyde (o-NBA) and 8 other aldehydes and ketones at glassy carbon cathodes in dimethylformamide containing various tetraalkylammonium tetrafluoroborate salts along with a proton donor (4-chlorophenol). Cyclic voltammograms for reduction of o-NBA exhibit three cathodic peaks attributable in succession to (a) one-electron generation of the nitro radical-anion, (b) three-electron formation of the hydroxylamine, and (c) two-electron production of benzisoxazole (anthranil). These findings have been employed to develop efficient controlled-potential (bulk) electrosyntheses of the following compounds: benzisoxazole, methylbenzo[c]isoxazole, [1,3]dioxolo[4 ‘,5 ‘,4,5]benzo[1,2-c]isoxazole, naphtho[2,3-c]isoxazole, 6-chlorobenzo[c]isoxazole, 6-methoxybenzo[c]isoxazole, 3-methyl-benzo[c]isoxazole, 3-isopropylbenzo[c]isoxazole, and 3-phenylbenzo[c]isoxazole. In addition, we have examined the use of a variety of proton donors to optimize the production of the desired product, and we have been able to recover the proton donor at the conclusion of the electrosynthesis. In each case, the synthesized product was separated by means of normal phase chromatography and identified with the aid of NMR spectros-copy, gas chromatography (GC), and gas chromatography-mass spectrometry (GC-MS). Isolated yields of the desired products range from 63 to 92 %. Moreover, our electrosyntheses are catalyst-free, environmentally green, and rapid (similar to 30 min).

Successful treatment of six patients with neuroleptic malignant syndrome associated with myoglobulinemic acute renal failure

Neuroleptic malignant syndrome is a rare but potentially lethal, rare reaction to neuroleptics which is characterized by altered levels of consciousness, extrapyramidal effects, autonomic instability, hyperthermia, and elevated serum creatine phosphokinase levels. The most serious complication of neuroleptic malignant syndrome is acute renal failure. We investigated six cases of neuroleptic malignant syndrome associated with myoglobulinemic acute renal failure due to rhabdomyolysis and effect of hemodialysis or hemodiafiltration. The patients were five males and one female with a mean age of 43.5 yr. All of the patients, who developed acute renal failure induced from rhabdomyolysis, had previously received butyrophenone (haloperidol), phenothiazine, benzamide, iminomide, benzisoxazole, antidepressants, and hypnotics (benzodiazepine and barbiturate) for the treatment of schizophrenia. The clinical manifestations of neuroleptic malignant syndrome were characterized by altered consciousness, muscle rigidity and weakness, fever, and excessive perspiration. The peak laboratory data were blood urea nitrogen 102 26 (mean SD) mg/dL, serum creatinine 9.1 2.1 mg/dL, serum creatine phosphokinase 229,720 289,940 IU/L, and all of them developed oliguric acute renal failure. Dantrolene sodium administration was given to five cases and hemodialysis or hemodiafiltration was performed in all of them. The serum creatinine level after hemodialysis or hemodiafiltration was 1.4 1.0 mg/dL. All patients were successfully cured of acute renal failure by hemodialysis or hemodiafiltration. As a result, myoglobulinemic acute renal failure associated with neuroleptic malignant syndrome was successfully treated by hemodialysis or hemodiafiltration.

Self-assembly in mixtures of sodium alkyl sulfates and alkyltrimethylammonium bromides: Aggregation behavior and catalytic properties

Two aqueous mixtures of cationic and anionic surfactants have been studied by means of conductometry, transmission electron microscopy, and microcalorimetry. Their catalytic effects on the decarboxylation of the kinetic probe 6-nitrobenzisoxazole-3-carboxylate (6-NBIC) were also examined in some detail. The mixtures differ profoundly in the hydrophobic match between both surfactant tails, which is perfect for dodecyltrimethylammonium bromide (DTAB) and sodium dodecyl sulfate (SDS) and poor for hexadecyltrimethylammonium bromide (CTAB) and sodium heptyl sulfate(SHS). This difference is reflected in the more pronounced synergism in critical aggregation concentration and catalytic efficiency of the DTAB/SDS mixture and in the phase behavior of the mixtures. CTAB and SHS can be mixed in a 1:1 ratio without precipitation, forming both small, unilamellar and large, multilamellar vesicles. In DTAB/SDS mixtures, however, precipitation of the catanionic surfactant occurs for a mole fraction of DTAB (x) between 0.3 and 0.7, while both vesicles and large bilayer fragments are formed for x = 0.8. The excess of DTAB in the x = 0.8 mixture results in the solubilization of the Vesicles by DTAB micelles close to the cmc of pure DTAB. A network of interconnected threadlike cylindrical micelles was found as an intermediate stage of aggregation between the vesicles and the mixed micelles. These cylindrical micelles are formed exclusively on further increasing the surfactant concentration.

Genotyping as a Key Element of Sample Size Optimization in Bioequivalence of Risperidone Tablets

Risperidone is a derivative of benzisoxazole and is widely used for schizophrenia and other psychiatric illnesses in both adults and children. Previous studies have confirmed that it is a highly variable drug (within-subject variability >= 30%). To reduce the large sample size required for bioequivalence researches on highly variable drugs, a role for genotyping in the design of the bioequivalence study was employed. A randomized, open-label, two-period crossover study was adopted: 20 subjects with specific genotypes carrying cytochrome P450 (CYP) 2D6*10 were randomized to two groups to receive a single oral dose of trial formulation or reference formulation with a 2-week washout period. Blood concentrations of risperidone (parent drug) and 9-hydroxy risperidone (active metabolite) were measured by high-performance liquid chromatography-tandem mass spectrometry. Eighteen out of the 20 subjects completed the study (two did not finish the test in the second period). The pharmacokinetic parameters of AUC(last), AUC(a) and C (max) for the 18 subjects after a single oral dose of the trial or reference preparation were 216.1 +/- 88.7 and 220.5 +/- 96.8 ng center dot h/mL; 221.6 +/- 93.1 and 226.4 +/- 103.5 ng center dot h/mL; 36.7 +/- 10.3 and 36.0 +/- 10.2 ng/mL, respectively. The CVw of risperidone in natural logarithm-transformed C (max) was 22.4 and 25.38% for 9-hydroxy risperidone. The test formulation met the Food and Drug Administration guidelines and regulation criteria for bioequivalence. By controlling the genotype, it could actually help reduce the CVw, which may be a feasible method to decrease the sample size for the bioequivalence study of highly variable drugs.

The decarboxylation of benzisoxazole-3-carboxylate has been investigated in detail by ab initio molecular orbital calculations. The effects of solvent on transition state geometries have been investigated by inclusion of one or two water molecules in the ab initio calculations. The decarboxylation and ring opening steps are found to be concerted. Kinetic isotope effects have been calculated for the carboxylate-C-13-labeled compound for various transition state geometries. Satisfactory agreement has been found between the experimental values for the reaction in water and ab initio HF/6-31G* calculated values for systems including four hydrogen bonds to the carboxylate group. The variations in free energies of solvation along the reaction path in five different solvents (water, methanol, chloroform, acetonitrile, tetrahydrofuran) have been calculated with Monte-Carlo free energy perturbation calculations. Solvent effects are generally overestimated, but the experimental trends have been reproduced for four of the five solvents, The effects of ion pairing have been tested by inclusion of a tetramethylguanidinium cation into the Monte-Carlo simulations for acetonitrile and tetrahydrofuran. With inclusion of ion pairing, the relative rates of THF and acetonitrile are reproduced much better, but solvent effects are underestimated relative to the reaction in water.

Synthesis and evaluation of radioiodine-labelled CP-118,954 for the in-vivo imaging of acetylcholinesterase

Objectives Alzheimer’s disease (AD) is characterized by reduced acetylcholinesterase (AChE) activity in the postmortem tissues of AD patients. Therefore, AChE has been an attractive target for the diagnosis of AD. In the present study, 5,7-dihydro-3-[2-(1-(phenyimethyl)-4piperidinyl)ethyl]-6H-pyrrolo[3,2-f]-1,2-benzisoxazol-6-one (CP-118,954), a potent AChE inhibitor, was labelled with radiolodine and evaluated as an AChE imaging agent for SPECT. Methods Radioiodine-labelled CP-118,954 was prepared from CP-144,885 and [I-125]iodobenzyl bromide, and anti-AChE activities of iodine-substituted CP-118,954 were measured. Metabolism studies were carried out in samples of blood and whole brain of mice injected with 2[I-123]iodo-CP-118,954 (I-123-1). Tissue distribution studies were also performed in mice injected with I-125-1, and samples of blood, thyroid, stomach, and brain tissue (cerebellum, striatum and cortex) were removed, weighed and counted. Results Of the ligands, 2-iodo-CP-118,954 exhibited higher binding affinity for AChE (IC50 = 24 nM) than the other positional isomers. 2-[I-125]lodo-CP-118,954 was found to have a lipophilicity (log P=2.1) favouring brain permeability and metabolic stability in mouse brain, but a marginal target (striatum) to non-target (cerebellum) uptake ratio (1.1) in mouse brain. Conclusion This result demonstrates that 2-[I-125]iodo-CP-118,954 may be unsuitable for AChE imaging. These findings suggest that radioligands suitable for AChE imaging should have not only a specific structure but also a sub-nanomolar to low nanomolar IC50.

Microwave-promoted synthesis of 3-amino-substituted 1,2-benzisoxazoles

Background: 1,2-benzisoxazole derivatives have been the focus of numerous studies, due to their biological and chemical interest. Results: We demonstrate an efficient synthesis of a series of 3-amino-substituted 1,2-benzisoxazoles from a 3-chloro-1,2-benzisoxazole by microwave-promoted nucleophilic aromatic substitution. The 3-amino-1,2-benzisoxazoles prepared were obtained in 1-6 h in good-to-high yields of 54-90%. The 3-chloro-1,2-benzisoxazoles were also prepared by heating with microwave irradiation in quantitative yields in 2 h, from the corresponding 3-hydroxy-1,2-benzisoxazoles. Conclusion: This efficient microwave-assisted pathway could be applied to a variety of substrates in the further development of substituted 1,2-benzisoxazoles.
