EXTRACTIVE SPECTROPHOTOMETRIC DETERMINATION OF QUETIAPINE FUMARATE IN PHARMACEUTICALS AND HUMAN URINE USING CALMAGITE AS AN ION-PAIR REAGENT

Quetiapine fumarate (QTF) is an antipsychotic drug belonging to the benzisoxazole derivatives indicated for the treatment of schizophrenia. A sensitive and selective method based on dichloromethane-extractable ion-pair of QTF with calmagite (CGT), which exhibited an absorption maximum at 490 nm, is described. At this. wavelength, Beer’s law is obeyed over the concentration range of 3.0-30.0 mu g ml(1). The apparent molar absorptivity, limit of detection (L OD) and quantitation (LOQ) values are 1.32×10(4) I mol(1) cm(-1), 0.27 and 0.81 mu g ml(1), respectively. The reaction is extremely rapid at room temperature and the absorbance values remain unchanged up to 19 h. The precision results, expressed as intra-day and inter-day relative standard deviation values, are satisfactory (RSD <= 2.2%). The accuracy is satisfactory as well (RE <= 2.44%). The method was successfully applied to the determination of QTF in pharmaceuticals and spiked human urine with satisfactory results. No interference was observed from common pharmaceutical adjuvants in tablets. Statistical comparison of the results with the official method showed an excellent agreement and indicated no significant difference in precision. Effect of anti-inflammatory bowel disease drug, E3040, on urate transport in rat renal brush border membrane vesicles

To confirm the assumption that 6-hydroxy-5,7-dimethyl-2-methylamino-4-(3-pyridylmethyl)benzothiazole (E3040) acts on urate reabsorption by inhibiting urate-anion exchange at the luminal membrane of renal tubules, we investigated the inhibitory effect of E3040 and its two conjugated metabolites on hydroxyl ion (OH-) gradient-dependent urate uptake into brush border membrane vesicles from rat renal cortex and compared it with other uricosuric agents. The order of potency was AA193 (5-chloro-7,8-dihydro-3-phenylfuro[2,3-g]-1,2-benzisoxazole-7-carboxylic acid)> benzbromarone > E3040 > probenecid > E3040 sulfate > E3040 glucuronide. Synthesis of novel 6-fluoro-3-(4-piperidinyl)-1,2-benzisoxazole derivatives as antiproliferative agents: A structure-activity relationship study

A series of novel 6-fluoro-3-(4-piperidinyl)-1,2-benzisoxazole derivatives 5(a-m) were synthesized with different substituted aromatic/heterocyclic acid chlorides (R-CO-Cl) and characterized by H-1 NMR, LC/MS, FTIR and elemental analyses. All the compounds synthesised were evaluated for their antiproliferative activity by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. The antiproliferative effects of the synthesised compounds were tested against viable human skin fibroblast cells and carcinoma cells namely HeLa cells, HT-29 cells, MCF-7 cells, HepG-2 cells by adopting positive and negative control. The importance of the aromatic and heterocyclic moiety was confirmed. From the SAR studies, it reveals that, the substitution at N-terminal of 6-fluoro-3-(4-piperidinyl)-1,2-benzisoxazole by the heterocyclic ring plays a dominant role and was responsible for the antiproliferative activity. Among the synthesized compounds 5a, 5d and 5k have showed potent antiproliferative activity on all the carcinoma cells tested.

1 Zonisamide, an anticonvulsant developed in Japan, is structurally similar to serotonin. Zonisamide has been proven to have a pharmacological profile that is very similar to that of carbamazepine. Hapten design for antibody-catalyzed decarboxylation and ring-opening reactions of benzisoxazoles

Three benzisoxazole haptens designed to elicit antibody binding sites with widely differing polarity have been synthesized and used to induce antibodies in mice. Monoclonal antibodies were prepared using hybridoma technology, and screened for catalysis of the ring-opening isomerization and/or decarboxylation of a series of related benzisoxazoles and their 3-carboxy-derivatives. No catalysis of decarboxylation was observed, but 3 of a total of 47 antibodies obtained against the three haptens catalyzed the isomerization process. Of 12 antibodies raised against the 3-acetylbenzisoxazole structure 5 none was catalytically active; but one of 24 raised against a 3-isopropenylbenzisoxazole 6 increased the rate of ring-opening of 6-nitrobenzisoxazole, and 5 of 11 antibodies raised against a benzisoxazole 7 with a 3-amidinium group were moderately active against either 6-nitro or 6-acylaminobenzisoxazoles. Competitive binding studies suggest that at least some of the antibodies induced by the isopropenyl hapten do possess a recognizably hydrophobic binding site.

Surface-enhanced Raman scattering and density functional theoretical study of anthranil adsorbed on colloidal silver particles

Surface-enhanced Raman spectrum of anthranil (2,1-benzisoxazole) in activated silver colloid was recorded and compared with the conventional Raman spectrum. The experimentally observed Raman bands along with their corresponding infrared bands were assigned based on the results of density functional theory (DFT) calculations. The significant changes evidenced between the SER and normal Raman spectra combined with the theoretical data obtained for Ag-anthranil model systems demonstrated that this molecule is adsorbed on the colloidal silver particles through the lone pair electrons of the nitrogen atom. The contribution of the chemical mechanism to the SERS enhancement was proved by the behavior of the electronic absorption spectrum of the mixture of activated silver colloid and anthranil solution. The orientation of the adsorbed species with respect to the metal surface was also predicted.

A redox-mediated Kemp eliminase

The acid/base-catalysed Kemp elimination of 5-nitro-benzisoxazole forming 2-cyano-4-nitrophenol has long served as a design platform of enzymes with non-natural reactions, providing new mechanistic insights in protein science. Here we describe an alternative concept based on redox catalysis by P450-BM3, leading to the same Kemp product via a fundamentally different mechanism. QM/MM computations show that it involves coordination of the substrate’s N-atom to haem-Fe(II) with electron transfer and concomitant N-O heterolysis liberating an intermediate having a nitrogen radical moiety Fe(III)-N-center dot and a phenoxyl anion. Product formation occurs by bond rotation and H-transfer. Two rationally chosen point mutations cause a notable increase in activity. The results shed light on the prevailing mechanistic uncertainties in human P450-catalysed metabolism of the immunomodulatory drug leflunomide, which likewise undergoes redox-mediated Kemp elimination by P450-BM3. Other isoxazole-based pharmaceuticals are probably also metabolized by a redox mechanism. Our work provides a basis for designing future artificial enzymes.

The latent photochromism of DNBP, as shown by x-ray analysis of the structures of the side-products and ESR/IR measurements, is attributed to open-shell reactions that are initiated by hydrogen photoabstraction and subsequent creation of two monoradicals, NH. and OH.. Large amounts of the radicals (ca 50% NH. and 70% OH) confined in the crystalline interior are persistent under ambient conditions. Through quasi-periodic reactions, the remaining radicals partially recover the ground-state isomers CH, NH and OH, or decay to the side-products, which results in crystalline photofatigue. Together with proton tunneling from the excited CH, the radical reactions represent dominant mechanism for the creation of NH and OH in the low-temperature regimes, but are successfully competed by the closed-shell reactions at higher temperatures. The precursor state, whose existence was assumed previously from transient absorption spectroscopy, may be identified as the radical OH.. The present work represents the first study of the photofatigue of a 2-(2,4-dinitrobenzyl)pyridine compound and extends the ‘classical’ mechanism of the photochromic reactions of nitrobenzylpyridines with a set of open-shell radical reaction routes.
