Catalysis of benzisoxazole isomerization by molecularly imprinted polymers

A tailor-made catalytically active polymer catalyzing the benzisoxazole isomerization is described. Kinetic studies carried out in water/ethanol (3:1, v/v) at room temperature, showed a rate acceleration (k(MIP)/k(control)) of 7.2-fold compared to the control polymer. The imprinted polymer exhibits Michaelis-Menten kinetics with a K-m of 0.484 mM and a k(cat) of 0.205 min(-1). Compared with the uncatalyzed reaction, a rate enhancement ((k(cat)/K-m)/k(uncat)) of 4 x 10(4) fold was obtained. Synthesis and evaluation of radioiodine-labelled CP-118,954 for the in-vivo imaging of acetylcholinesterase

Objectives Alzheimer’s disease (AD) is characterized by reduced acetylcholinesterase (AChE) activity in the postmortem tissues of AD patients. Therefore, AChE has been an attractive target for the diagnosis of AD. In the present study, 5,7-dihydro-3-[2-(1-(phenyimethyl)-4piperidinyl)ethyl]-6H-pyrrolo[3,2-f]-1,2-benzisoxazol-6-one (CP-118,954), a potent AChE inhibitor, was labelled with radiolodine and evaluated as an AChE imaging agent for SPECT. Methods Radioiodine-labelled CP-118,954 was prepared from CP-144,885 and [I-125]iodobenzyl bromide, and anti-AChE activities of iodine-substituted CP-118,954 were measured. Metabolism studies were carried out in samples of blood and whole brain of mice injected with 2[I-123]iodo-CP-118,954 (I-123-1). Tissue distribution studies were also performed in mice injected with I-125-1, and samples of blood, thyroid, stomach, and brain tissue (cerebellum, striatum and cortex) were removed, weighed and counted. Results Of the ligands, 2-iodo-CP-118,954 exhibited higher binding affinity for AChE (IC50 = 24 nM) than the other positional isomers. 2-[I-125]lodo-CP-118,954 was found to have a lipophilicity (log P=2.1) favouring brain permeability and metabolic stability in mouse brain, but a marginal target (striatum) to non-target (cerebellum) uptake ratio (1.1) in mouse brain. Conclusion This result demonstrates that 2-[I-125]iodo-CP-118,954 may be unsuitable for AChE imaging. These findings suggest that radioligands suitable for AChE imaging should have not only a specific structure but also a sub-nanomolar to low nanomolar IC50.

Safety and Efficacy of Zonisamide in Refractory Epilepsy Patients: Clinical Experience from a Tertiary Center

Objectives: Zonisamide (ZnS) is a new generation antiepileptic agent used in the treatment of epilepsy patients with partial and generalized seizures. In this study, we aim to investigate the safety and efficacy of ZnS in the treatment of patients with refractory epilepsy who were being followed. Methods: Forty-five refractory epilepsy patients who received ZnS treatment were included in this study. Patients who received ZnS treatment for less than six months were excluded. Age, sex, types of seizures, examination findings, magnetic resonance imaging and electroencephalography findings, concurrent use of non-ZnS antiepileptic drugs, decrease in the seizure frequency and side effects of the drug were recorded. Results: Thirty-nine patients, whose mean age was 34.3 +/- 9.3 years, were evaluated. Complex partial seizures (CPS) and generalized tonic-clonic seizure (GTCS) were observed in 74.4% of the patients, whereas 10.3% had GTCS alone, 7.7% had CPS alone, 5.2% had GTCS and myoclonia and 2.6% of them had absence and myoclonia. In the follow-up, treatment was observed to be discontinued in 19 of the 39 patients due to drug side effects, or where there was an increase, or no change, in seizure frequency. Twenty patients responded to treatment. Seizure frequency was decreased by 25% in one patient; 50% in five patients, and 75% in seven patients. Three patients were seizure-free. Although there was no change in seizure frequency, seizure duration was shortened in four patients. Treatment-responsive patients were using ZnS at doses that ranged from 100 to 400 mg/day for 7 to 80 months. Conclusion: ZnS is a safe, tolerable and effective option for the additional treatment of refractory epilepsy patients at our center.

Photochemistry of matrix-isolated 3-chloro-1,2-benzisoxazole: Generation and characterization of 2-cyanophenoxyl radical and other reactive intermediates

Photochemistry of 3-chloro-1,2-benzisoxazole 1 in N-2 and Ar matrices at 10 K leads to N-chloro-ketenimine 3 and 2-cyanophenyl-hypochlorite 4. The reaction kinetics and the observed photoisomerization of 3 to 4 indicate that ketenimine 3, possibly formed via an elusive vinylnitrene VN, is an intermediate in the formation of hypochlorite 4. A new pathway involving the formation of 2-cyanophenoxyl radical 5, which was captured only in Ar matrix, was also observed. Radical 5 is possibly formed via photo detachment of CI atom from I (or VN) and might explain the formation of 3-chloro-6-oxocyclohexa-1,4-dienecarbonitrile 2 in N2 and Ar matrices. All the species were characterized by IR spectroscopy and theoretical calculations. The computed geometric and electronic structure of radical 5 is discussed. Overall, the results provided further insight into the mechanism of the photochemistry of 1,2-benzisoxazoles and allowed characterization of new interesting reactive intermediates. Synthesis and evaluation of amide, sulfonamide and urea – benzisoxazole derivatives as potential atypical antipsychotics

In this paper, we report the optimization of a series of novel, potential antipsychotic derivatives combining potent dopamine D-2, D-3 and serotonin 5-HT1A, 5-HT2A receptor affinities. The pharmacological features of compound 27 are a high affinity for dopamine D-2, D-3 and serotonin 5-HT1A, 5-HT2A receptors. Clinical pharmacology and therapeutic drug monitoring of zonisamide

Zonisamide (1,2-benzisoxazole-3-methanesulfonamide) is a new antiepileptic drug developed in Japan. This compound is insoluble in water, and it is available in tablet and powder form. In experimental animals, this compound has been found to have a strong inhibitory effect on convulsions of cortical origin because it suppresses focal spiking and the spread of secondary generalized seizures. In humans, a series of double-blind, placebo-controlled studies revealed the efficacy of zonisamide for patients with refractory partial seizures and for selected patients with infantile spasms. Its antiepileptic mechanism of action remains unclear, but it is likely to involve blockade of both sodium and T-type calcium channels. Oral bioavailability of zonisamide is excellent in healthy human volunteers. Zonisamide is slowly absorbed and has a mean t(max) of 5 to 6 hours. Almost 100% of it is absorbed; there is no difference in bioavailability between tablets and powder. Zonisamide concentrations are highest in erythrocytes and then in whole blood and plasma. It is approximately 40% to 60% bound to plasma proteins, primarily albumin. Its volume distribution is 0.9 to 1.4 L/kg. In adults, the elimination half-life is between 50 and 62 hours, and it takes as long as 2 weeks to reach steady state. The dose-serum level correlation is linear up to doses of 10 to 15 mg/kg per day, and the therapeutic range is 10 to 40 mu g/ml. However, the relationship between serum zonisamide levels, clinical response, and adverse effects appears weak. Concurrent enzyme-inducing anticonvulsants such as phenytoin, carbamazepine, or barbiturates stimulate zonisamide metabolism and decrease serum zonisamide levels at steady state. Although zonisamide has been reported to increase the serum levels of phenytoin and carbamazepine in some patients, the interactions of zonisamide with other antiepileptic drugs seem to be of minor clinical relevance. A pilot study of zonisamide suppositories revealed that it is beneficial for patients with neurologic disorders in whom antiepileptic drugs cannot be administered by mouth.
