To a solution of the above product (1.0 g, 3.5 mmol) in DMF (15 ml) was added NaOMe solution in methanol (30% wt/wt, 10 ml). The mixture was heated to 8O0C under vacuum to remove residual methanol and then stirred at 11O0C under nitrogen overnight. The mixture was then cooled to room temperature, diluted with water (50 ml), adjusted to pH 6 with 10% aqueous HCl. The resulting suspension was stirred in an ice bath for 2 hours and filtered. The solid was washed with water and dried in an oven (900C) for 2 hours and then under high vacuum at room temperature for 3 hours to obtain a slightly yellow solid. LC-MS m/e: (M+ 1) = 282

The yield was 72% based on Compound (X-1) as the starting material. The obtained crystal was pale brown. 1H NMR (deuterated DMSO): delta6.95-6.98 (1H), delta7.02-7.04 (1H), delta7.40-7.51 (3H), delta7.90-7.95 (1H), delta8.75 (1H), delta8.85-8.88 (2H), delta9.03 (2H), delta10.89 (1H).

Herein, we have designed various benzisoxazole acetamide derivatives with and without glycine spacer as DPP-IV inhibitors. Compounds 9a?d and 11a?e were synthesized and screened for their in vitro DPP-IV inhibition. Compounds 11a and 11c showed moderate activity for DPP-IV inhibition, whereas other remained inactive at 25?200 muM concentrations. DPP-IV inhibition can be a good strategy for modulating diabetes and cancer; hence, we have screened compounds 9a?d and 11a?e for their anticancer activity using MTT assay against A549 and MCF7 cell lines. Compounds 9a?d without glycine spacer have shown good anticancer activity compared to compounds 11a?e with glycine spacer. Compound 9b has shown moderate activity with IC50 values 4.72 ¡À 0.72 and 4.39 ¡À 0.809 muM against A549 and MCF7 cell lines, respectively. Interestingly, compound 9c with cyano group has shown very good anticancer activity with IC50 2.36 ¡À 0.34 muM against MCF7 cell line as compared to fluorouracil with IC50 45.04 ¡À 1.02 muM.

The reaction kinetics and the observed photoisomerization of 3 to 4 indicate that ketenimine 3, possibly formed via an elusive vinylnitrene VN, is an intermediate in the formation of hypochlorite 4. A new pathway involving the formation of 2-cyanophenoxyl radical 5, which was captured only in Ar matrix, was also observed. Radical 5 is possibly formed via photodetachment of Cl atom from 1 (or VN) and might explain the formation of 3-chloro-6-oxocyclohexa-1,4-dienecarbonitrile 2 in N2 and Ar matrices. All the species were characterized by IR spectroscopy and theoretical calculations. The computed geometric and electronic structure of radical 5 is discussed. Overall, the results provided further insight into the mechanism of the photochemistry of 1,2-benzisoxazoles and allowed characterization of new interesting reactive intermediates.
