However, when the incubation between enzyme and inhibitor was prolonged to 1d h, the obtained K-1 was of 35.2 nM, of the same order of magnitude as that of the clinically used sulfonamides/sulfamates acetazolantide, methazolamide, ethoxzolamide and topiramate (K(1)s in the range of 5.4-15.4 nM). The inhibition of the human mitochondrial isozyme hCA V with these compounds has been also tested by means of a dansylamide competition binding assay, which showed zonisamide and topiramate to be effective inhibitors, with K(1)s in the range of 20.6-25.4 nM. The X-ray crystal structure of the adduct of hCA 11 with zonisamide has also been solved at a resolution of 1.70 A, showing that the sulfonamide moiety participates in the classical interactions with the Zn(II) ion and the residues Thr199 and Glu106, whereas the benzisoxazole ring, is oriented toward the hydrophobic half of the active site, establishing a large number of strong van der Waals interactions (< 4.5 A) with residues Gln92, Vall2l, Phe131, Leu198, Thr200, Pro202. Au(I)-catalyzed coupling of benzisoxazoles and ynamides was recently reported to synthesize challenging indoloquinoline core structures. In this report, we employed DFT methods to elucidate the mechanistic details of this reaction. The results reveal that the catalytic cycle involves initial coupling of ynamide with benzisoxazole, simultaneous ring opening of the isoxazole unit and attack of the dimethoxybenzene (DMOB) unit to gold carbenoid, proton transfer from the DMOB group to hydroxyl group, thioether migration, ring closure, and proton transfer. The experimentally observed ligand-controlled product selectivity is reproduced well by the calculations. The product selectivity-determining step is the ring opening of the isoxazole unit. The flexible P(t-Bu)(2)(o-biphenyl) ligand facilitates the approach of the DMOB group to gold carbenoid, which brings about significant C-H/pi interactions in the transition state for ring opening, and thus leads to the indoloquinoline product. The rigid 1,3-bis(diisopropylphenyl)imidazol-2-ylidene ligand prefers keeping the DMOB group away from the gold carbenoid, which can cause strong orbital interaction in the transition state for ring opening, and thus results in indole product.

The sulfide derivatives (2a-f) were obtained by the nucleophilic substitution of bromo derivatives (1a-f) with thiophenols, and sulfone derivatives (3a-f) were obtained by the oxidation of 2a-f. All the newly synthesized compounds were characterized by elemental analysis, infrared, nuclear magnetic resonance, and mass spectroscopic data. Furthermore, these novel sulfide and sulfone derivatives were screened for their antibacterial and antifungal activities against various microorganisms.

Acetylcholinesterase (AChE) is an important cholinergic marker of Alzheimer’s disease (AD) and shows reduced activity in postmortem AD brain tissues. 1-(4-Fluorobenzyl)-4-[(5,6-dimethoxy-l-oxoindan-2-fluoro-2-yl)methyl]piperidine (G379, 1), an AChE inhibitor with a subnanomolar IC50 (0.56 nM), was prepared as a F-18-labeled radioligand ([F-18]1) and evaluated in mice. Metabolism studies of [F-18]1 showed no metabolites in the mouse brain. Tissue distribution studies demonstrated its uniform regional distribution in the mouse brain, suggesting that this radioligand is not suitable for the in vivo imaging of AChE. This result along with reports on radiolabeled AT-benzylpiperidine lactam benzisoxazole (IC50 < 1 nM) and other radiolabeled benzylpiperidme derivatives (IC50 > 1 nM) suggested that a subnanornolar IC50 may not be the only important factor in determining the suitability of a radioligand for in Vivo Studies of AChE. Novel derivatives of 6-fluoro-4-piperidinyl-1,2-benzisoxazole amides 4(I-VI) were obtained by the condensation of different acid chlorides with 6-fluoro-3-piperidin-4yl-benzo[d]isoxazole. Also, 6-fluoro-chroman-2-carboxamides 6(I-III) were synthesized by using nebulic acid chloride with different amines in presence of triethylamine as acid scavenger and dichloroethane as solvent. The synthesized compounds were characterized by IR, H-1 NMR, and CHN analysis. These molecules were evaluated for their efficacy as antimicrobials in vitro by disc diffusion and microdilution method against pathogenic strains such as Bacillus substilis, Escherichia coli, Pseudomonas fluorescens, Xanthomonas campestris pus, X. oryzae, Aspergillus niger, A. flavus, Fusarium oxysporum, Trichoderma species, F monaliforme, and Penicillum species. Compounds 4I, 4IV, 4V, 1I, 6II and 6III showed better inhibitory activity than compared to standard drugs. Among these compounds, 4IV and 6III showed potent inhibitory activity against all the strains and found to be nonstrain dependent.
