EFFECT OF NOVEL BENZISOXAZOLE DERIVATIVES AGAINST EHRLICH ASCITES CARCINOMA CELLS IN SWISS ALBINO MICE: CYTOTOXIC AND HAEMATALOGICAL STUDIES

The present study is to investigate the cytotoxicity properties and haematological indices of synthesized molecules (S1-S4) in comparison to the standard drug 5 fluorouracil which reflects the anticancer potentials of the above molecules. Cytotoxicity studies were performed by in vitro MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl teterazolium bromide) assay. Haematological studies were done by drawing blood in the drug and synthesized molecules administered animals by retroorbital method. Haemoglobin, RBC, WBC, lymphocytes, Monocytes and Granulocytes counts were measured. All the above experiments were performed against Ehrlich ascites carcinoma (EAC) in Swiss albino mice. The percentage cytotoxicity and percentage viable cell counts revealed anticancer properties of the synthesized molecules. RBC count indicated protection of cell membrane from destruction and the WBC counts along with lymphocytes, Monocytes and granulocytes also show the immunoprotective characteristics of the novel molecules. Thus it has been concluded that the synthesized molecules (S1-S4) if supported by further molecular studies may have a promising role to play as anticancer agents.

CTAB and SHS can be mixed in a 1:1 ratio without precipitation, forming both small, unilamellar and large, multilamellar vesicles. In DTAB/SDS mixtures, however, precipitation of the catanionic surfactant occurs for a mole fraction of DTAB (x) between 0.3 and 0.7, while both vesicles and large bilayer fragments are formed for x = 0.8. The excess of DTAB in the x = 0.8 mixture results in the solubilization of the Vesicles by DTAB micelles close to the cmc of pure DTAB. A network of interconnected threadlike cylindrical micelles was found as an intermediate stage of aggregation between the vesicles and the mixed micelles. These cylindrical micelles are formed exclusively on further increasing the surfactant concentration.

SYNTHESIS AND CATALYTIC PROPERTIES OF CROSS-LINKED HYDROPHOBICALLY ASSOCIATING POLY(ALKYLMETHYLDIALLYLAMMONIUM BROMIDES)

Cross-linked, hydrophobically associating homo- and copolymers were synthesized by free-radical cyclo(co)polymerization of alkylmethyldiallylammonium bromide monomers with a small amount of N,N’-methylenebisacrylamide in aqueous solution using ammonium persulfate as the initiator. The hydrophobic microdomains of the cross-linked polysoaps were characterized by hypsochromic shifts of the long-wavelength absorption band of methyl orange as a solvatochromic probe, noncovalently bound to the macromolecule. Catalysis of the unimolecular decarboxylation of 6-nitrobenzisoxazole-3-carboxylate by the cross-linked copolymers was investigated in aqueous solution at pH 11.3 and 30 degrees C. The cross-linked polysoaps exhibited higher catalytic activities for decarboxylation than the corresponding non-cross-linked copolymer analogues. A maximum in rate constant was found at about 0.2% (w/w) of cross-linking agent in the cross-linked copolymers.

Multi-pharmacophore Approach to Bio-therapeutics: Piperazine Bridged Pseudo-peptidic Urea/Thiourea Derivatives as Anti-oxidant Agents

In an attempt to develop synergetic, potential and selective anti-oxidant agents, a series of novel piperazine bridged pseudo-peptidic urea/thiourea derivatives (5a-x) are designed by molecular docking study, synthesized and evaluated for their in-vitro anti-oxidant activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and dimethyl-4-phenylenediamine (DMPD) methods. The results revealed that compounds 5q, 5s, 5w and 5x are good anti-oxidant agents. Amongst, compounds with aromatic side chain, thioureal fragment with electron donating group-substitution, particularly, 5s and 5w are the most active members exhibiting remarkable anti-oxidant property. The docking results are complementary to the experimental results. The correlation study revealed that compounds derived from phenylalanine and tryptophan containing electron donating group are excellent antioxidants. [GRAPHICS] .

Delineation of the Role of Astroglial GABA Transporters in Seizure Control

Studies of GABA transport in neurons and astrocytes have provided evidence that termination of GABA as neurotransmitter is brought about primarily by active transport into the presynaptic, GABAergic nerve endings. NOVEL BENZISOXAZOLE DERIVATIVES AS POTENT AND SELECTIVE INHIBITORS OF ACETYLCHOLINESTERASE

A series of N-benzylpiperidine benzisoxazoles has been developed as potent and selective inhibitors of the enzyme acetylcholinesterase (AChE). The benzisoxazole heterocycle was found to be an appropriate bioisosteric replacement for the benzoyl functionality present in the N-benzylpiperidine class of inhibitors. The title compounds were synthesized by alkylating 3-methyl-1,2-benzisoxazoles with an iodo piperidine derivative as the key step. Benzisoxazoles 1b-j,o displayed potent inhibition of AChE in vitro with IC50’s = 0.8-14 nM. Particularly interesting were N-acetyl and morpholino derivatives Ig (IC50 = 3 nM) and 1j (IC50 = 0.8 nM), respectively, which displayed outstanding selectivity for acetyl- over butyrylcholinesterase, in excess of 3 orders of magnitude. N-Acetyl 1g also displayed a favorable profile in vivo. This analog showed a dose-dependent elevation of total acetylcholine in mouse forebrain after oral administration with an ED(50) = 2.4 mg/kg. In addition, 1g was able to reverse amnesia in a mouse passive avoidance model at doses of 3.2 and 5.6 mg/kg with an average reversal of 89.7%. Molecular dynamics simulations were used to study the possible binding modes of N-benzylpiperidine benzisoxazoles to AChE from Torpedo californica, Key structural insights were obtained regarding the potency of this class of inhibitors. Specifically, Asp-72, Trp-84, Trp-279, Phe-288, and Phe-330 are implicated in the binding of these inhibitors. The N-benzylpiperidine benzisoxazoles may be suitable compounds for the palliative treatment of Alzheimer’s Disease.

The 5-HT2A antagonist MDL-100,907 fully generalized to S-16924 and two further 5-HT2A antagonists, fananserin and SR-46349, showed partial generalization. However, MDL-100,907, fananserin, and SR-46349 showed less pronounced generalization to clozapine. Similarly, the 5-HT2C antagonists SB-200,646 and SB-206,553 more markedly generalized to S-16924 than to clozapine. The 5-HT1A receptor agonist (+/-)-8-dihydroxy-2-(di-n-propylamino) tetralin generalized fully to S-16924 but not to clozapine. Full generalization was obtained to both S-16924 and clozapine for the clozapine congeners, olanzapine and quetiapine. In distinction, the benzisoxazole, risperidone, and the phenylindole, sertindole, weakly generalized to S-16924 and clozapine. However, the benzisoxazole ziprasidone, which possesses 5-HT1A agonist properties, generalized fully to S-16924 but not to clozapine. 2,1-Benzisoxazoles have been selectively synthesized through reductive heterocyclization of 2-nitroacylarenes using Pt supported nanoparticles. The reaction involves a cascade process in which the first step is the reduction of the nitro group into hydroxylamine followed by heterocyclization through the nucleophilic attack of the hydroxylamine group to the carbonyl of the acyl group and further dehydration. The reaction was performed on Pt/C, Pt/TiO2, and Pt/MgO using hydrogen as the reducing agent under mild reaction conditions. The results showed that Pt/MgO was the most active and selective catalyst. The study of the influence of the crystal size of the metal on the activity and selectivity, combined with the reaction mechanism examined by in situ Fourier transform infrared spectroscopy of the adsorbed reactant, showed that the maximum activity and selectivity to the target compound can be achieved by controlling the architecture of metal particles and acid base properties of the support. The effect of temperature on selectivity, the stability of the Pt/MgO catalyst, and the scope of the reaction have been studied. Finally, reductive heterocyclization using different metals (Pd and Au) supported on MgO has also been performed.

Synthesis and antimicrobial activity of 3-propene 1,2-benzisoxazole derivatives

A convenient and eco-friendly synthesis of 3-propene 1,2-benzisoxazole derivatives 5a-h has been achieved in excellent yields from the corresponding substituted o-hydroxy acetophenones. Substituted o-hydroxy acetophenones 1a-h on treatment with acetaldehyde 2 in sodium hydroxide afford 1-(2′-hydroxy aryl)-2-butene-1-ones 3a-h. Compounds 3a-h on oximation yields 4a-h. Microwave irradiation of 4a-h on solid support silica gel affords 5a-h in very good yield. The resultant 3-propene 1,2-benzisoxazoles have been characterized by spectral data. The compounds 5a-h have been screened for their antimicrobial activity. Compounds 5b, d, e, g exhibit good antibacterial activity and compounds 5b-e exhibit market antifungal activity.

Paliperidone in the treatment of delirium: results of a prospective open-label pilot trial

Objective: Delirium is a life-threatening neuropsychiatric syndrome characterised by disturbances in consciousness, attention, cognition and perception. Antipsychotics are considered the drugs of choice in managing the symptoms of delirium. Paliperidone is a benzisoxazole derivative and the principal active metabolite of risperidone. In this study, we aimed to evaluate the efficacy of paliperidone for the treatment of delirium. Methods: A prospective open-label study of paliperidone for delirium treatment was performed with 6-day follow-up. Fifteen patients who met Diagnostic and Statistical Manual of Mental disorders, Fourth Edition criteria for delirium and had a score of 13 on the Delirium Rating Scale were recruited. The starting dose was 3 mg once a day and the dose was adjusted depending on the status of delirium. Daily assessments of the severity of delirium were evaluated using Memorial Delirium Assessment Scale (MDAS). Results: The mean daily maintenance dose of paliperidone was 3.75 +/- 1.06. The MDAS scores before and after treatment (day 7) were 23.60 +/- 6.31 and 11.33 +/- 5.45 (t = 6.78, p < 0.001), respectively. The intensity of delirium showed a statistically significant reduction in MDAS scores from the first day of treatment. No serious adverse effects were observed, and none of the patients discontinued paliperidone because of adverse effects. Conclusions: This study shows that low-dose paliperidone is effective in reducing behavioural disturbances and symptoms in delirium and is well tolerated in delirious patients. This trial is an open-label study with a small sample size, and further controlled studies will be necessary.
