Cytokinin-like activity of N ‘-substituted N-phenylureas

We have synthesized 14 N-phenylurea derivatives, differing in the heterocyclic portion linked in N’-position, and tested their cytokinin-like activity. Three different bioassays were used: the chlorophyll level determination test, the bioassay for the expression of hormone-induced chimeric Pg5-GUS gene and the tomato regeneration test, in which 1,2-benzisoxazole-3-acetic acid (BOAA) was utilized as auxin. The cytokinin-like activity showed by three of these compounds in the regeneration assay seems to be related to their different heterocyclic nature. Results obtained indicate that the N-phenyl-N’-1,3,4-thiadiazol-2-ylurea (compound 4), an isomer of N-phenyl-N’- 1,2,3-thiadiazol-5-ylurea (thidiazuron, TDZ), in the absence of auxin induces shoot regeneration in the 34,2% of the explants cultured; the N-plienyl-N’-(3-chloro-1,2-benzisothiazol-7-yl) urea (compound 10), structurally different from TDZ, in the absence of auxin induces shoot regeneration in the 25,9% of explants, significantly lower than that of TDZ (68,8%). N-phenyl-N’-benzothiazol-6-ylurea (compound 13), structurally different from TDZ, in the absence of auxin induces the 99,5% of shoot regeneration, significantly different from that of the other substances. The addition of auxin in the cotyledon regeneration assay reduces the differences. The compound 13 could be considered a new phenylurea derivative with a highly specific cytokinin-like activity.

Synthesis, characterization and evaluation of antioxidant and anticancer activities of novel benzisoxazole-substituted-allyl derivatives

A novel series of various 2-allylbenzo[d]isoxazol-3(2H)-ones were synthesized using benzo[d]isoxazol-3(2H)-one treated with different allyl bromides/chlorides in the presence of water-mediated cesium carbonate as a new catalyst 3(a-h). Biotransformation of the drug occurred through oxidative N-dealkylation (formation of the acid metabolite M1), monohydroxylation of the alicyclic ring (M9), alcohol dehydrogenation (formation of the ketone metabolite M12), and benzisoxazole scission (formation of M11), the latter in combination with glucuronidation (M16) or alicyclic hydroxylation (M10). Unchanged drug, M1, M9, M12, and M16 were detected in urine; M10 and M11 were detected in feces. The monohydroxylated metabolite M9 was solely present in urine samples of extensive CYP2D6 metabolizers, whereas M10, another metabolite monohydroxylated at the alicyclic ring system, was present in feces of poor metabolizers as well. Nboc decarboxylation rates increased in the presence of vesicles prepared with both phospholipids and DODAC/phospholipid mixtures. Quantitative analysis of vesicular effects was done using pseudophase models. Phospholipids catalyzed up to 140-fold while the maximum catalysis by DODAC/lipid vesicles reached 800-fold. Acceleration depends on alkyl chain length, fatty acid insaturation of the lipids, and the DODAC/phospholipid molar ratio. Catalysis is not related to the liquid crystalline-gel state of the bilayer and may be related to the relative position of Nboc with respect to the interface.

Their interaction with aqueous medium in biological systems may be simulated by considering their interaction with explicit water molecules. Such studies provide information on the structures, energies and type of interactions stabilizing the resulting geometric systems. The objective of the current study was to utilize theoretical approaches to investigate the structures, stabilization energy and binding energy of benzox-azole-water, 1,2-benzisoxazole-water and 2,1-benzisoxazole-water complexes. The calculations were performed utilizing the density functional theory (DFT)/M06-2X/6-311++G(d, p) method and the DFT/omega B97XD method with both the 6-311++G(d, p) and the aug-cc-pVDZ basis sets. The results suggest that the stability of the different clusters depends on interrelated factors including the rings formed by intermolecular hydrogen bonds and the proton affinity (PA) or acidity of the atoms forming the intermolecular hydrogen bonds with the water molecules. A comparison across methods indicates that the results follow similar trends with different methods.
