Overview of Japanese experience-controlled and uncontrolled trials

Zonisamide is a new type of benzisoxazole derivative, first marketed in Japan in 1989. This study analyzed: (1) the drug’s efficacy by seizure and epilepsy type in a total of 1008 patients treated during the development of zonisamide in Japan; (2) the effectiveness of zonisamide for 726 newly-diagnosed patients treated with zonisamide postmarketing; and (3) 50 patients with generalized epilepsies and epileptic syndromes (idiopathic generalized epilepsies, symptomatic generalized epilepsies, Lennox-Gastaut syndrome, Doose syndrome, and West syndrome), and 19 patients with undetermined epilepsies and specific syndromes (refractory grand mat in childhood, severe myoclonic epilepsy in infancy, other undetermined epilepsy, familial. essential myoclonic epilepsy, and mitochondrial encephalomyopathy with ragged-red fibers). Analysis of study results showed that among all patients treated, zonisamide was highly effective for the treatment of idiopathic generalized epilepsy, temporal lobe epilepsy, and other partial epilepsies. The compound was also effective for other symptomatic generalized epilepsies. In 50 patients with generalized epilepsies, and 19 with undetermined epilepsies and specific syndromes, seizure frequency was reduced by >50% with monotherapy or two-drug therapy with zonisamide. Zonisamide is a benzisoxazole-based compound first synthesized in the early 1970s by the research laboratories of Dainippon Pharmaceutical Company in Osaka, Japan. Identified as an anticonvulsant during exploratory research, zonisamide has since been characterized as having broad-spectrum antiepitepsy and neuroprotective effects. Effects of zonisamide on dopaminergic system

Effects of zonisamide (ZNS) on extracellular dopamine (DA), its precursor 3,4-dihydroxyphenylalanine (DOPA), its metabolites 3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) levels in the striatum as well as hippocampus of freely moving rats were studied. Intracellular DA, DOPA, DOPAC and HVA levels, as well as DOPA accumulation as an index of tyrosine hydroxylase activity in the rat brain in vivo, DA re-uptake in the striatum and hippocampus, and monoamine oxidase (MAO) activities were also determined. Acute administrations of therapeutic ZNS doses (20 and 50 mg/kg) increased striatal extracellular DOPA levels, intracellular striatal and hippocampal DOPA levels, and stimulated DOPA accumulation in both brain regions. ZNS also increased striatal and hippocampal intracellular as well as extracellular DA and MIA Levels, but decreased those of DOPAC levels. Chronic (3 weeks) administrations of therapeutic ZNS doses (20 and 50 mg/kg/day) increased intracellular DA, DOPA, DOPAC and HVA levels in striatum and hippocampus. ZNS-induced changes were greater in intracellular levels than in extracellular levels. Acute and chronic supratherapeutic ZNS dose (100 mg/kg) administration decreased intracellular levels of all substances detectable in both brain regions, and inhibited DOPA accumulation. Both subtypes of MAO (type A and type B) activities were weakly inhibited by ZNS. ZNS showed no effect on DA re-uptake in striatum nor in hippocampus. These results suggest that therapeutic ZNS doses increase DOPA accumulation as well as both intracellular and extracellular DA, DOPA and HVA levels. However, such doses also decrease extracellular and intracellular DOPAC levels by enhancing DA synthesis and/or by selectively inhibiting MAO-B activities. In addition, chronic therapeutic ZNS dose administration enhances DA synthesis, which results in increased intracellular DA, its precursor and its metabolites levels. On the other hand, both acute and chronic supratherapeutic ZNS dose administrations inhibit DA turnover. These ZNS effects on DA metabolism are at least partly involved in the mechanisms of action of ZNS.

Coordination Cages Based on Bis(pyrazolylpyridine) Ligands: Structures, Dynamic Behavior, Guest Binding, and Catalysis

CONSPECTUS: We describe here a family of coordination cages with interesting structural, guest-binding, and catalytic properties. Flexible bridging ligands containing two bidentate pyrazolylpyridine termini assemble with transition-metal dications to afford coordination cages containing a metal ion at each vertex, a bridging ligand spanning each edge, and a 2:3 metal:ligand ratio. This stoichiometry is expressed in structures ranging from M4L6 tetrahedra to M16L24 tetracapped truncated tetrahedra, which are stabilized by the formation of pi-stacked arrays between electron-rich and electron-poor ligand segments that form around the cage periphery. In some cases concentration- and/or temperature-dependent equilibria between multiple cage structures occur, arising from a balance between entropy, which favors the formation of a larger number of smaller assemblies, and enthalpy, which maximizes both interligand aromatic stacking and solvophobic effects in the larger assembles. The cages are hollow and can accommodate guests often anions or solvent molecules in the central cavity. For one cage family, M8L12 species with an approximately cubic structure and a ca. 400 angstrom(3) cavity, the guest binding properties have been studied extensively. This cage can accommodate a wide range of neutral organic guests, with binding in water being driven principally by the hydrophobic effect, which leads to binding constants of up to 10(8) W-1. The accumulation of a large amount of empirical data on guest binding in the M8L12 cage in water provided the basis for a predictive tool for in silico screening of potential guests using the molecular docking program GOLD; this methodology has allowed the identification of numerous new guests with accurately predicted binding constants and provides a transformative new approach to exploring the host/guest chemistry of cages. Binding of benzisoxazole inside the M8L12 cage results in substantial rate enhancements by a factor of up to 2 x 10(3) -of the Kemp elimination, in which benzisoxazole reacts to give 2-cyanophenolate. Catalysis arises because the 16+ cage cation accumulates anions around the surface by ion pairing, leading to a high effective concentration of hydroxide ions surrounding the guest even when the bulk pH is modest. Thus, the catalysis relies on the operation of two orthogonal interactions that bring the reaction partners together: hydrophobic guest binding in the cavity, which is lined with CH groups from the ligands, and ion pairing around the highly cationic cage surface. A consequence of this is that under some conditions the product of the cage catalyzed Kemp elimination (the 2-cyanophenolate anion) itself accumulates around the cage surface and deprotonates another benzisoxazole guest, perpetuating the reaction in an autocatalytic manner. Thus, different anions accumulating around the cage can act as partners for reaction with a cavity-bound guest, opening up the possibility that the M8L12 cage can act as a general catalyst for reactions of electrophilic guests with surface-bound anions.

Solvolysis of Tris-p-nitrophenyl-phosphate in aqueous and reverse micelles

The widespread use of toxic phosphates and phosphonates as insecticides, and their use as chemical weapons, has led to investigation of fast detoxification and decontamination methods. Micelles, microemulsions, cyclodextrines and liposomes have been used to accelerate phosphate ester decomposition by nucleophiles. Here, hydrolysis, methanolysis and hexanolysis of Tris-p-nitrophenyl phosphate (TNPP), a model for reactive phosphate esters. were studied in homogeneous phase, aqueous and reverse micelles. Kinetic micellar effects were quantitatively analyzed using pseudo-phase models. TNPP hydrolysis was catalyzed by cetyltrimethylammonium chloride (CTAC), cetyltrimethylammonium bromide (CTAB). and hexadecylammonium propanesulfonate (HPS), micelles by factors of five, CTAC, and three. CTAB, HPS, respectively. The calculated rate constants for spontaneous and acetate-catalyzed hydrolysis in the micellar phase were significantly higher than those in the aqueous phase. While in water and in methanol the effect of the acetate cation was negligible, the catalytic efficiency of acetate for hexanolysis depended on the nature of the cation with the K+ salt being ca. 20 times more efficient than the tetraethylammonium salt in non-polar solvents. Sodium dodecylsulfate, SDS. micelles inhibited TNPP hydrolysis by a factor of eigth. Reverse micelles of CTAB in n-hexanol/isooctane (10:90, v/v) did not catalyze TNPP hydrolysis, but changed the bis-p-nitrophenyl phosphate/hexyl-bis-p-nitrophenylphosphate product ratio depending of CTAB concentration and water/detergent ratio.
