HDAC and NF-kappa B mediated cytotoxicity induced by novel N-Chloro beta-lactams and benzisoxazole derivatives

Novel N-chloro a-Lactam and benzisoxazole derivatives were successfully synthesized with excellent yields (92-96%) under simple and mild reaction conditions. The beta-lactams as a class acquired importance since the discovery of penicillin which contains beta-lactam unit as an essential structural feature of its molecule, this interest continued unabated because of the therapeutic importance of beta-lactam antibiotics. In silico studies of the compounds with cancer drug target enzymes showed the inhibition of HDAC (Histone Deacetylase) and NF-kappa B (nuclear factor kappa-light-chain-enhancer of activated B cells) significantly. The compounds were then investigated for the inhibitory potential against the same enzymes in vitro. NF-kappa B inhibition was investigated by trans activation assay using HEK293/NF-kappa B-luc cells. Overall, the synthesized compounds induce the cancer cell toxicity by restraining the NF-kappa B transcription factor mediated by HDAC inhibition and thus the compounds act as dual inhibitors. Cocrystals of zonisamide: physicochemical characterization and sustained release solid forms

We report four cocrystals of the antiepileptic drug, zonisamide (ZNS), which encounters half-life fluctuation when administered adjunctly with other antiepileptic drugs. Single crystals for two of the novel cocrystals of ZNS were successfully prepared from solvent evaporation experiments and their crystal structures were determined. Pharmaceutically acceptable cocrystals were analyzed for their dissolution rate, solubility and stability to draw conclusions on the impact of cocrystallization on the physicochemical properties of ZNS. It was found that the cocrystals showed lower solubility and dissolution rates and offer potential benefits in the development of sustained release formulations of ZNS which could address issues regarding its half-life fluctuation. Recent attempts to explore newer therapeutic applications have suggested ZNS as a potential drug for weight loss management. In this regard, the cocrystal of ZNS with caffeine, which has also been used in weight loss management, promises potential applications in the development of a novel fixed-dose combination drug which could offer synergistic therapeutic benefits in the treatment of obesity.

Risperidone is an approved antipsychotic drug belonging to the chemical class of benzisoxazole. This drug has low solubility in aqueous medium and poor bioavailability due to extensive first-pass metabolism and high protein binding (>90%). As new strategies to improve treatments efficiency are needed, we have studied cationic G4 PAMAM dendrimers’ performance to act as efficient nanocarriers for this therapeutic drug. In this respect, we explored dendrimer risperidone complexation dependence on solvent, temperature, pH and salt concentration, as well as in vitro cytotoxicity measured on L929 cell line and human red blood cells. The best dendrimer risperidone incorporation was achieved when a mixture of 70:30 and 90:10 v/v chloroform:methanol was used, obtaining 17 and 32 risperidone molecules per dendrimer, respectively. No cytotoxicity on L929 cells was found when dendrimer concentration was below 3 x 10(-2) mu M and risperidone concentration below 5.1 mu M. Also, no significant hemolysis or morphological changes were observed on human red blood cells. Finally, attempting to obtain an efficient drug delivery system for risperidone, incorporation in G4 PAMAM dendrimers was optimized, improving drug solubility with low cytotoxicity. The stability of two benzisoxazole antipsychotics was determined in vitro in decomposing porcine blood inoculated with bacteria, utilizing a high-performance liquid chromatography with ultraviolet and fluorescence detection method for drug quantitation. Stability experiments for risperidone and paliperidone were conducted at 7, 20 and 37 degrees C for 4 days using sterile and bacterially inoculated porcine blood. The benzisoxazole-cleaved degradation products for both drugs were identified as 2-hydroxybenzoyl-risperidone and 2-hydroxybenzoyl-paliperidone utilizing liquid chromatography quadrupole-time-of-flight mass spectrometry and accurate mass measurements. The degradation products have been found in postmortem case studies, including one case where risperidone and paliperidone were not detected, indicating complete conversion can occur in situ.

AN AUTOMATED PROCEDURE FOR SIMULATING CHEMICAL-REACTIONS IN SOLUTION – APPLICATION TO THE DECARBOXYLATION OF 3-CARBOXYBENZISOXAZOLE IN WATER

A combined Monte Carlo quantum mechanical and molecular mechanical (QM/MM) simulation method is described for the investigation of the solvent effects on chemical reactions, In the present approach, ab initio molecular orbital calculations are first used to locate the transition state, from which the reaction path is determined by using Gaussian 90. Then, free energy changes between adjacent structures generated along this intrinsic reaction path are evaluated via statistical perturbation theory using the combined QM/MM-AM1/TIP3P potential. Since empirical parametrization of the reaction system is not needed in these calculations, the method presented here is essentially an automated procedure for simulating reactions in solution, which may be conveniently used by organic chemists. We have employed the procedure to examine the decarboxylation of 3-carboxybenzisoxazole in aqueous solution. The predicted free energy of activation is 26.1 +/- 0.3 kcal/mol, in excellent agreement with the experimental value of 26.3 kcal/mol. Analyses of the contributing factors in solute-solvent interaction suggest that the aqueous solvent effect is primarily due to the difference in the intrinsic (in vacuo) charge distributions between the reactant and transition state. Solvent polarization contributes significantly to the solute-solvent interaction; however, the nature of the electronic polarization of the reactant and the transition state is markedly different.

The envelope proteins VP26 and VP28 play a major role in infection process and also in the interaction with the host cells. A comprehensive study on the viral proteins leading to the development of safe and potent antiviral therapeutic is of adverse need. The novel synthesized compound 3-(1-chloropiperidin-4-yl)-6-fluoro benzisoxazole 2 is proved to have potent antiviral activity against WSSV. The compound antiviral activity is validated in freshwater crabs (Paratelphusa hydrodomous). An in silico molecular docking and simulation analysis of the envelope proteins VP26 and VP28 with the ligand 3-(1-chloropiperidin-4-yl)-6-fluoro benzisoxazole 2 are carried out. The docking analysis reveals that the polar amino acids in the pore region of the envelope proteins were involved in the ligand binding. The influence of the ligand binding on the proteins is validated by the molecular dynamics and simulation study. These in silico approaches together demonstrate the ligand’s efficiency in preventing the trimers from exhibiting their physiological function.
