Novel DNA Gyrase Inhibiting Spiropyrimidinetriones with a Benzisoxazole Scaffold: SAR and in Vivo Characterization

The compounds described herein with a spirocyclic architecture fused to a benzisoxazole ring represent a new class of antibacterial agents that operate by inhibition of DNA gyrase as corroborated in an enzyme assay and by the inhibition of precursor thymidine into DNA during cell growth. A high drug exposure NOEL on oral dosing in the rat suggested that a high therapeutic margin could be achieved. Design, synthesis and anticancer activity of functionalized spiro-quinolines with barbituric and thiobarbituric acids

A new series of spiro-quinoline compounds have been accomplished by the reaction of barbituric acid or thiobarbituric acid with derivatives of benzisoxazole-5-carbaldehyde or 2-substituted benzaldehyde. These compounds were evaluated for their in vitro cytotoxicity on two mammalian cancer cell lines MCF-7 and KB. The compounds exhibit cytotoxicity against these cell lines in micromolar range. The decarboxylation of benzisoxazole-3-carboxylate has been investigated in detail by ab initio molecular orbital calculations. The effects of solvent on transition state geometries have been investigated by inclusion of one or two water molecules in the ab initio calculations. The decarboxylation and ring opening steps are found to be concerted. Kinetic isotope effects have been calculated for the carboxylate-C-13-labeled compound for various transition state geometries. Satisfactory agreement has been found between the experimental values for the reaction in water and ab initio HF/6-31G* calculated values for systems including four hydrogen bonds to the carboxylate group. The variations in free energies of solvation along the reaction path in five different solvents (water, methanol, chloroform, acetonitrile, tetrahydrofuran) have been calculated with Monte-Carlo free energy perturbation calculations. Solvent effects are generally overestimated, but the experimental trends have been reproduced for four of the five solvents, The effects of ion pairing have been tested by inclusion of a tetramethylguanidinium cation into the Monte-Carlo simulations for acetonitrile and tetrahydrofuran. With inclusion of ion pairing, the relative rates of THF and acetonitrile are reproduced much better, but solvent effects are underestimated relative to the reaction in water.

The inhibition of the human mitochondrial isozyme hCA V with these compounds has been also tested by means of a dansylamide competition binding assay, which showed zonisamide and topiramate to be effective inhibitors, with K(1)s in the range of 20.6-25.4 nM. The X-ray crystal structure of the adduct of hCA 11 with zonisamide has also been solved at a resolution of 1.70 A, showing that the sulfonamide moiety participates in the classical interactions with the Zn(II) ion and the residues Thr199 and Glu106, whereas the benzisoxazole ring, is oriented toward the hydrophobic half of the active site, establishing a large number of strong van der Waals interactions (< 4.5 A) with residues Gln92, Vall2l, Phe131, Leu198, Thr200, Pro202. An unusual case of risperidone instability in a fatality presenting an analytical and interpretative challenge

This paper describes the detection and characterization of unusual metabolite/breakdown products of the anti-psychotic drug risperidone in post-mortem blood and urine as part of a toxicological investigation into an unexpected death of a male suffering from paranoid schizophrenia prescribed risperidone and previously paroxetine. Compounds detected in the post-mortem blood and urine specimens were shown to be benzisoxazole ring scission products of risperidone and a hydroxy metabolite. These compounds are never routinely detected in blood and urine but can be present in mammalian faeces indicating that gut bacteria could be responsible for their formation. In this case, evidence for this process was demonstrated by the controlled in vitro stability study of risperidone spiked into the case blood and urine leading to the hypothesis that the post-mortem blood and urine samples analyzed could have contained bacteria with the ability to breakdown risperidone and its metabolite in this way. This finding is very unusual and has not been encountered before in any previous risperidone cases investigated by the authors, or widely reported in the post-mortem toxicological literature. However, a recently published paper has supported these findings in blood. As a result of this work, it was shown that the deceased had taken risperidone prior to death, even in the absence of any risperidone or its hydroxy metabolite(s) in the blood and urine.
