SYNTHESIS AND CATALYTIC PROPERTIES OF HYDROPHOBICALLY-MODIFIED POLY(ALKYLMETHYLDIALLYLAMMONIUM CHLORIDES)

Novel non-cross-linked and cross-linked, hydrophobically modified homo- and copolymers were synthesized by free-radical cyclo(co)polymerization of alkylmethyldiallylammonium chloride monomers in aqueous solution using ammonium persulfate as the initiator. Cross-linking was brought about by addition of a small amount of N,N’-methylenebisacrylamide. The cross-linked homo- and copolymers showed an increase of their reduced viscosity in aqueous solution upon the controlled introduction of cross-linking agent into their chemical structure. Viscosity measurements revealed that the conformational transition of polysoaps to compact coils in aqueous solution is strongly dependent upon the hydrophobic group content of the polysoaps. The formation of hydrophobic microdomains is akin to intramolecular micelle formation. Depending on the hydrophobic group content and the percentage of cross-linking, intermolecular aggregation was also revealed by viscosity measurements at higher concentrations of polysoap. The hydrophobic microdomains of the non-cross-linked and cross-linked polysoaps were characterized by hypsochromic shifts of the long-wavelength absorption band of Methyl Orange as a solvatochromic probe, non-covalently bound to the macromolecule. Catalysis of the unimolecular decarboxylation of 6-nitrobenzisoxazole-3-carboxylate by the non-cross-linked and crosslinked copolymers was investigated in aqueous solution at pH 11.3 and 30 degrees C. The cross-linked polysoaps exhibited higher catalytic activities for decarboxylation than the corresponding non-cross-linked analogues. A maximum in rate constant was found at about 0.2% (w/w) of cross-linking agent in the cross-linked polysoaps. The decarboxylation rate is strongly dependent upon the hydrophobic group content in the polysoaps.

However, when the incubation between enzyme and inhibitor was prolonged to 1d h, the obtained K-1 was of 35.2 nM, of the same order of magnitude as that of the clinically used sulfonamides/sulfamates acetazolantide, methazolamide, ethoxzolamide and topiramate (K(1)s in the range of 5.4-15.4 nM). The inhibition of the human mitochondrial isozyme hCA V with these compounds has been also tested by means of a dansylamide competition binding assay, which showed zonisamide and topiramate to be effective inhibitors, with K(1)s in the range of 20.6-25.4 nM. The X-ray crystal structure of the adduct of hCA 11 with zonisamide has also been solved at a resolution of 1.70 A, showing that the sulfonamide moiety participates in the classical interactions with the Zn(II) ion and the residues Thr199 and Glu106, whereas the benzisoxazole ring, is oriented toward the hydrophobic half of the active site, establishing a large number of strong van der Waals interactions (< 4.5 A) with residues Gln92, Vall2l, Phe131, Leu198, Thr200, Pro202. Photochemistry of matrix-isolated 3-chloro-1,2-benzisoxazole: Generation and characterization of 2-cyanophenoxyl radical and other reactive intermediates

Photochemistry of 3-chloro-1,2-benzisoxazole 1 in N-2 and Ar matrices at 10 K leads to N-chloro-ketenimine 3 and 2-cyanophenyl-hypochlorite 4. The reaction kinetics and the observed photoisomerization of 3 to 4 indicate that ketenimine 3, possibly formed via an elusive vinylnitrene VN, is an intermediate in the formation of hypochlorite 4. A new pathway involving the formation of 2-cyanophenoxyl radical 5, which was captured only in Ar matrix, was also observed. Radical 5 is possibly formed via photo detachment of CI atom from I (or VN) and might explain the formation of 3-chloro-6-oxocyclohexa-1,4-dienecarbonitrile 2 in N2 and Ar matrices. All the species were characterized by IR spectroscopy and theoretical calculations. The computed geometric and electronic structure of radical 5 is discussed. Overall, the results provided further insight into the mechanism of the photochemistry of 1,2-benzisoxazoles and allowed characterization of new interesting reactive intermediates. 1 Zonisamide, an anticonvulsant developed in Japan, is structurally similar to serotonin. Zonisamide has been proven to have a pharmacological profile that is very similar to that of carbamazepine. A practical synthesis of benzisoxazole 1 and its conversion to alpha-aryloxyisobutyric acid 2 using 1,1,1-trichloro-2-methyl-2-propanol (chloretone) was developed. Benzisoxazole I was formed in high yields by the action of either methanesulfonyl chloride/base upon intermediate oxime 8 or with thionyl chloride/base, which initially forms cyclic sulfite 10. A highly reactive, short-lived intermediate derived from chloretone was detected by ReacIR and its half-life determined to be similar to 5 min. Reaction conditions for the Bargellini reaction were developed that resulted in a 95% yield of 2 from the reaction of highly hindered phenol 1 with chloretone hemihydrate and powdered NaOH in acetone. Thus highly hindered alpha-aryloxyisobutyric acids can be made in a single step in high yield.

In silico docking studies and synthesis of new phosphoramidate derivatives of 6-fluoro-3-(piperidin-4-yl)benzo[d]isoxazole as potential antimicrobial agents

A new class of phosphoramidate derivatives of 6-fluoro-3-(piperidin-4-yl)benzo[d]isoxazole were synthesized in good to excellent yields (78-96%) by an in situ, three-step process. All the synthesized molecules were evaluated for anti-bacterial and anti-fungal activities using in vitro and in silico methods. The results revealed that the compounds 4b, 4d, 4h, 4i, and 4j exhibited the most promising anti-bacterial activity against S. aureus, B. subtilis, K. pneumoniae, S. typhi and P. mirabilis and anti-fungal activity against A. niger and A. flavus when compared with the standard drugs Norfloxacin and Nystatin at concentrations of 25, 50, 75 and 100 mu g/mL. The rest of the title compounds have shown moderate activity against all the bacterial and fungal strains. Molecular docking studies revealed that the synthesized compounds have exhibited significant binding modes with high dock scores ranging from -7.2 to -9.5 against 3V2B protein when compared with the standard drugs Norfloxacin (-5.8) and Nystatin (-6.6) respectively. Hence, it is suggested that the synthesized phosphoramidate derivatives of 6-fluoro-3-(piperidin-4-yl)benzo[d]isoxazole will stand as the promising antimicrobial drug candidates in future.

These micelles and those of the corresponding carboxybetaines are more effective catalysts than those of the corresponding cationic surfactants, In all cases a change from Me to Pr at the head group speeds reaction by factors of ca. 5-8 for the sulfobetaines and amine oxides and ca. 14 for the cationic surfactants. Cyclizations of the o-3-halopropyloxyphenoxide ions (halogen = Br, I), which are intramolecular S(N)2 reactions, are modestly micellar catalysed, but structural effects on the micellar catalysis by cationic and betaine surfactants are in the same sequence, as for decarboxylation.

Catalysis of benzisoxazole isomerization by molecularly imprinted polymers

A tailor-made catalytically active polymer catalyzing the benzisoxazole isomerization is described. Kinetic studies carried out in water/ethanol (3:1, v/v) at room temperature, showed a rate acceleration (k(MIP)/k(control)) of 7.2-fold compared to the control polymer. The imprinted polymer exhibits Michaelis-Menten kinetics with a K-m of 0.484 mM and a k(cat) of 0.205 min(-1). Compared with the uncatalyzed reaction, a rate enhancement ((k(cat)/K-m)/k(uncat)) of 4 x 10(4) fold was obtained. Substrate selectivity, accessible binding site analysis, dissociation constant determination, and inhibition study were also performed.

Process improvements for the preparation of kilo quantities of a series of isoindoline compounds

A series of isoindoline analogues with either an indazole (HMR 2934, HMR 2651) or benzisoxazole (HMR 2543) appendage were prepared for the proposed treatment of psychiatric disorders such as obsessive compulsive disorder and attention deficit disorder.
