SYNTHESIS AND EVALUATION OF A SERIES OF 3,5-DISUBSTITUTED BENZISOXAZOLE-4,7-DIONES – POTENT RADIOSENSITIZERS INVITRO

A series of 3,5-disubstituted-2,1-benzisoxazole-4,7-diones was synthesized and evaluated as radiosensitizers both in vitro and in vivo. These compounds were designed as non-nitro electron-affinic agents in an effort to alleviate some of the toxicities seen with the 2-nitroimidazole radiosensitizers evaluated in the clinic. Several compounds in this series were potent radiosensitizers in vitro, with sensitizer enhancement ratios of 2.0-2.3 at concentrations < 0.5 mM. Compounds with potent in vitro activity were also evaluated in vivo. However, none of these compounds showed radiosensitizing activity in vivo. The reduction potentials of these compounds were determined by cyclic voltammetry and compared to other electron-affinic radiosensitizers. In general, the reduction potentials of this series of compounds was slightly more positive than the 2-nitroimidazoles, but they fell within the range postulated as acceptable to yield in vivo activity. The results suggest that factors other than reduction potential may be responsible for the lack of in vivo radiosensitizing activity observed for this class of radiosensitizers. The percentage of initiator used in the polymerisation, ranging from 1% to 3%, although it does not have a substantial effect on the catalytic rate, reduces considerably the imprinting efficiency. The alteration of the template/monomer ratio is also investigated, and the data show that there is considerable loss of imprinting efficiency. In terms of substrate selectivity, a number of experiments have been performed using 5-Cl-benzisoxazole as substrate analogue, as well as 5-nitro-indole as template analogue for the preparation of a different set of nanogels. All the kinetic data demonstrate that the chemical structure of the template is key to the molecular recognition properties of the imprinted nanogels that are closely tailored and able to differentiate among small structural changes. Zonisamide: A Review of Its Use in the Management of Adults with Partial Seizures

Oral zonisamide (Zonegran (R)) is a benzisoxazole derivative chemically unrelated to other antiepileptic agents. In a double-blind, multinational study in adults newly diagnosed with partial seizures, shorter-term monotherapy with once-daily zonisamide was noninferior to that with twice-daily carbamazepine controlled release in terms of seizure freedom according to the International League Against Epilepsy guidelines, with seizure freedom benefits maintained during longer-term therapy. In four randomized, double-blind, placebo-controlled studies in adults with refractory partial seizures, shorter-term adjunctive therapy with once-or twice-daily zonisamide reduced the frequency of seizures to a significantly greater extent than placebo, with antiepileptic efficacy sustained following longer-term treatment in this patient population. Zonisamide was generally well tolerated in adults with partial seizures participating in these studies, with the majority of adverse events being mild or moderate in severity. Thus, oral zonisamide as monotherapy or adjunctive therapy to other AEDs provides a useful option in the treatment of patients with partial seizures.

Surface-enhanced Raman scattering and density functional theoretical study of anthranil adsorbed on colloidal silver particles

Surface-enhanced Raman spectrum of anthranil (2,1-benzisoxazole) in activated silver colloid was recorded and compared with the conventional Raman spectrum. The experimentally observed Raman bands along with their corresponding infrared bands were assigned based on the results of density functional theory (DFT) calculations. The significant changes evidenced between the SER and normal Raman spectra combined with the theoretical data obtained for Ag-anthranil model systems demonstrated that this molecule is adsorbed on the colloidal silver particles through the lone pair electrons of the nitrogen atom. The contribution of the chemical mechanism to the SERS enhancement was proved by the behavior of the electronic absorption spectrum of the mixture of activated silver colloid and anthranil solution. The orientation of the adsorbed species with respect to the metal surface was also predicted.

Quetiapine fumarate (QTF) is an antipsychotic drug belonging to the benzisoxazole derivatives indicated for the treatment of schizophrenia. A sensitive and selective method based on dichloromethane-extractable ion-pair of QTF with calmagite (CGT), which exhibited an absorption maximum at 490 nm, is described. At this. wavelength, Beer’s law is obeyed over the concentration range of 3.0-30.0 mu g ml(1). The apparent molar absorptivity, limit of detection (L OD) and quantitation (LOQ) values are 1.32×10(4) I mol(1) cm(-1), 0.27 and 0.81 mu g ml(1), respectively. The reaction is extremely rapid at room temperature and the absorbance values remain unchanged up to 19 h. The precision results, expressed as intra-day and inter-day relative standard deviation values, are satisfactory (RSD <= 2.2%). The accuracy is satisfactory as well (RE <= 2.44%). The method was successfully applied to the determination of QTF in pharmaceuticals and spiked human urine with satisfactory results. No interference was observed from common pharmaceutical adjuvants in tablets. Statistical comparison of the results with the official method showed an excellent agreement and indicated no significant difference in precision.
